高温放线菌固体复合菌剂的制备及发酵条件的优化
Preparation and fermentation conditions optimization of thermophilic actinomycetes solid compound microbial agent

Abstract
Five Actinomycetes with high temperature tolerance and cellulose degradation ability were screened from compost samples at high temperature period,and prepared into solid composite microbial agents.The degradation ability and core strains of the compound microbial agent were determined by cellulose degrading enzyme activity assay and strain simplification experiment.Single factor and response surface methodology experiments were used to optimize the fermentation conditions of solid compound microbial agents.The results showed that the enzyme activities of filter paper and cellulase of the compound microbial agent were higher than those of the single strain,and reached the highest on the 13th day.Strain GW-8 was the core strain of the compound microbial agent.The optimal solid-state fermentation conditions were as follows:the carbon source was starch,the nitrogen source was peptone,the initial water content was 40%,the inoculum volume was 0.35 mL/g,The spore number of solid microbial agent can reach 1.63×108 CFU/g after 9 days of culture at 45℃.In the corn straw degradation experiment,the degradation rate of WB group added with solid microbial agent reached 30.83%,which was significantly higher than the other two groups,and the degradation rate increased by 78.41%compared with the treatment group without adding.The high-temperature actinomycetes solid compound microbial agent effectively improved the degradation rate of corn straw,which can be further applied to the high-temperature composting treatment and field returning treatment of corn straw.It provided technical support for the resource utilization of agricultural waste.关键词
