肠道微生物与长寿的因果关系及免疫细胞的中介作用:一项孟德尔随机化研究
Causal effect of gut microbiota on longevity and the mediating role of im-mune cells:a Mendelian randomized study
摘要
Abstract
Aim:To elucidate the causal effects of gut microbiota on longevity,and the mediating role of immune cells using Mendel randomized(MR)method.Methods:Genome-wide association study(GWAS)data of gut microbiota and im-mune cells were downloaded from https://www.ebi.ac.uk/gwas/downloads/summary-statistics,and those of longevity were from https://www.longevitygenomics.org/download.Single nucleotide polymorphisms(SNPs)significantly associated with gut microbiota were screened as instrumental variables at P<1×10-5.Inverse-variance weighted(IVW)method was used to assess the causal effect of gut microbiota on longevity,while mediation analysis was applied to explore the potential medi-ating role of immune cells.Results:Using IVW method,14 gut microbes were identified to be causally related with longevi-ty,and 9 were positively and 5 were negatively(P<0.05).33 immune cells straits significantly impacting on longevity were screened out,and 18 of which were positively correlated and 15 negatively correlated(P<0.05)13 immune cell traits were found to mediate the impact of gut microbiota on longevity,including CD20 on IgD+CD24-,CD20 on IgD+,IgD on unsw mem,CD3 on naive CD8br,PB/PC AC,Naive CD8br AC,CD3 on HLA DR+CD8br,FSC-A on monocyte,CD11c+mono-cyte% monocyte,CD20 on transitional,IgD-CD24-AC,CD39+CD8br% T cell,CD45RA on TD CD8br,the mediation effect proportion was-30.28%-18.89%.Conclusion:Gut microbiota could effect causally on longevity through immune cell-mediated mechanisms.关键词
