摘要

Abstract

Aim:To investigate effects of hypoxia on expression of hypoxia-inducible factor-2α(HIF-2α)and migra-tion,invasion abilities of esophageal squamous cell carcinoma(ESCC)cells.Methods:RT-qPCR and immunohistochemistry were used to detect the expression of HIF-2α in cancerous tissues and paracancerous normal esophageal tissues of 30 ESCC patients,esophageal normal epithelial cells(HEEC)and ESCC cells(KYSE150 and EC109).A cell culture hypoxic envi-ronment was constructed to detect the effect of hypoxia on the expression of HIF-2α in ESCC cells.Scratch and Transwell assays were used to detect the effects of hypoxia,knockdown of HIF-2α and knockdown of HIF-2α during hypoxia on ESCC cells'function.Results:The expression of HIF-2α elevated in ESCC tissues compared with paracancerous normal esophage-al tissues,and ESCC cells compared with HEEC(P<0.001).Hypoxia promoted the expression of HIF-2α,migration and invasion abilities of ESCC cells;knockdown of HIF-2α inhibited the expression of HIF-2α,migration and invasion abilities;hypoxia and knockdown of HIF-2α had an interactive effect(P<0.05).Conclusion:Hypoxia could regulate the progres-sion of ESCC by modulating the expression of HIF-2α in ESCC cells.
