益气养阴活血方对糖尿病肾病大鼠肾功能及SIRT3/SOD2/GPX4通路、铁死亡的影响
Effects of Yiqi Yangyin Huoxue recipe on renal function,SIRT3/SOD2/GPX4 pathway and ferroptosis in rats with diabetic nephropathy

Abstract
Aim:To explore the effects and possible mechanism of Yiqi Yangyin Huoxue recipe on renal function in rats with diabetic nephropathy(DN).Methods:Totally 42 rats were randomly allocated into normal control group(n=6)and the modeling group(n=36),and the rats in the modeling group were fed with high-fat and high-sugar chow and com-bined with a single intraperitoneal injection of streptozotocin to establish a DN model.The successfully constructed model rats(n=32)were allocated into the model group,the valsartan group(the positive control group),the low dose group of Yiqi Yangyin Huoxue recipe(the low-dose group),and the high-dose group,with 8 rats in each group.The rats in the latter 2 groups were administered with the Chinese traditional medicine at 11.0 and 22.0 g/kg respectively,and the gavage lasted for 6 weeks.After 24 hour urine and blood were collected,the rats were executed,and kidney tissues were taken.The deter-mination of 24 hour total urinary protein(24 h-UTP),serum creatinine(SCr),glucose(GLU),triglycerides(TG),albumin(ALB)by AU5800 Auto-biochemical Analyzer,serum alanine aminotransferase(ALT)was determined by the rate method,and the serum malondialdehyde(MDA)was detected by ELISA.The renal histopathological changes were observed by HE staining.The mRNA and protein expressions of renal tissue silent information regulator 3(SIRT3),superoxide dismutase 2(SOD2),glutathione peroxidase 4(GPX4),and solute carrier family 7 member 11(SLC7A11)were detected by real-time fluorescence quantitative PCR and Western blot.Results:Compared with the normal control group,rats in the model group had severe kidney pathological damage,higher 24 h-UTP,SCr,GLU,TG,ALT and MDA,and lower ALB,lower kidney SIRT3,SOD2,SLC7A11,and GPX4 mRNA and protein expression levels(P<0.05);compared with the model group,the pathological damage of the kidney was alleviated,the above indexes in the low and high dose groups were improved(P<0.05),and there was no significant difference in the improvements between the 2 groups(P>0.05).Conclusion:Yiqi Yangyin Huoxue recipe may inhibit oxidative stress and ferroptosis by activating SIRT3/SOD2/GPX4 signaling pathway,thus reduce renal damage in rats with DN.关键词
