Study on chromosome doubling induction and ploidy identification in potato true seeds

Abstract
[Objective]Potato is the most important tuber crop worldwide.Cultivated potatoes are autotetraploid,and their narrow genetic background leads to significant homogenization in hybrid breeding offspring.Therefore,breeding with diploid potatoes,which have a more diverse genetic background,is one of the new trends in potato breeding.Among them,using diploid potato true seeds as the subject and creating polyploids through chromosome doubling technology is an efficient method for developing new germplasm resources.[Method]In this study,diploid potato true seeds were taken as the research object,based on the principle that colchicine can induce chromosome doubling,the liquid immersion method and the solid medium method were used to explore the doubling effect of different treatment methods,treatment concentrations and treatment time on potato true seeds,thus identifying the polyploidization effect of different doubling methods.Morphological and cytological identification methods were used to identify the phenotype of potato materials before and after doubling,and the characteristics of the doubled plants were analyzed.[Result]The solid medium method could efficiently obtain doubled plants.The doubling rate of some genotypes could be as high as 91%,and the doubled plants had thick stalks and deepened leaf colour,which had the characteristics of polyploidy.The method can be directly applied to the solid seed,with high throughput,simple operation and time-saving.Compared with the classical colchicine dip method,the solid medium method could be widely applicable to a wide range of potato genotypes.Compared with the original diploid material,the agronomic traits of the doubled material were enhanced in terms of plant height,stem diameter,pollen size and other aspects.[Conclusion]This method adopts the direct doubling of potato true seeds,which has the characteristics of high efficiency,high throughput,simple operation and wide applicability,thus providing theoretical basis and technical support for the creation and research of potato tetraploid germplasm resources.关键词
