芥菜型油菜含油量不同检测方法的比较及重组自交系群体种子含油量的变异分析
Comparison of different detection methods for oil content and variation analysis of seed oil content of recombinant inbred lines of Brassica juncea

Abstract
[Objective]This study aims to compare the efficacy of three oil content detection methods and analyze the variation in seed oil content among recombinant inbred lines(RILs)of Brassica juncea.The research purpose is to provide resources for breeding high-oil-content Brassica juncea and to lay the foundation for quantitative trait locus(QTL)mapping of oil content.[Method]Oil content of 18 new lines of Brassica juncea was measured using nuclear magnetic resonance(NMR)spectroscopy,near-infrared(NIR)spectroscopy,and the residue method.The oil content of recombinant inbred lines under two environmental conditions(Xining and Huzhu)was determined using NMR spectroscopy.[Result]The results showed that the oil content measured by NIR spectroscopy was generally higher than that measured by the other two methods.The results of NMR spectroscopy and the residue method were similar,demonstrating higher stability and reliability.Significant variations in the oil content of the population were observed under both environmental conditions.In Xining in 2020,the oil content ranged from 26.69%to 45.80%,while in Huzhu in 2020,it ranged from 26.31%to 46.49%.Materials under the two environmental conditions were classified into three groups through cluster analysis,and elite breeding resources with high oil content were identified.[Conclusion]NMR spectroscopy performed best in practical applications.The efficiency of breeding selection for oil content traits was high,and high-oil-content breeding resources were identified.This study provides a theoretical foundation and practical guidance for breeding high-oil-content Brassica juncea.关键词
