Abstract
To evaluate the feasibility of the adenosine triphosphate(ATP)biofluorescence 24-h assay for rapid microbial detection in cosmetics and related fields,concerning the guiding principles for validation of microbial qualitative test methods of the Chinese Pharmacopoeia(2020 edition),six different types of cosmetics and eight representative strains were selected for investigation of the equivalence with the microbiological standard test methods in the Safety and Technical standards for Cosmetics(2015 edition)in terms of method specificity,detection limit,reproducibility and durability.The eight representative strains grew well in the dual-culture system,and the presence of the test samples had no significant effect on the results,indicating that the ATP amplified biofluorescence 24-h assay method has good specificity.The representative strains were inoculated with six cosmetic products at target concentrations of 10,1,and 0.1 CFU/g.The results showed that the detection limits of the ATP amplified biofluorescence 24-h assay were in the range of 0.28 CFU/g~1.69 CFU/g,which was significantly better than that of the standard assay(p<0.000 1).The results of the inter-laboratory comparison showed that there was no statistically significant difference between the ATP amplified biofluorescence 24-h assay and the standard method for the same product(p>0.05).Meanwhile,the results of the online detection after enrichment using a TLE medium with different pH values were positive,indicating that the method has good reproducibility and durability.Therefore,the ATP amplified biofluorescence 24-h assay is partially equivalent to the standard method and can be used for the rapid screening of microbial contaminated products in cosmetics.关键词
