Abstract

The DEAD-box gene family,the largest subfamily of RNA helicases,plays a crucial role in various aspects of biological genetic processes,including replication,repair,recombination,transcription,RNA transport,organelle gene expression and translation.Based on the genome data of melon,bioinformatics approaches were employed to screen and identify members of the DEAD-box family,and their physicochemical properties,chromosomal locations,phylogeny,gene structures,conserved motifs and differential expression at different stages of fruit development were analyzed.The results indicated that 29 CmDEADs genes were identified in melon,distributed across 12 chromosomes.The protein sequence lengths ranged from 409-1 172 aa,with isoelectric points(PI)spanning from 5.29-9.95,and molecular weights ranging from 46 430.54-129 660.04 Da.Phylogenetic analysis revealed that the 29 CmDEADs could be divided into four subgroups,with subgroups Ⅰ-Ⅳ containing 10,8,6,5 genes,respectively,and collinearity analysis showed that 11 melon CmDEADs had collinearity relationships with 16 genes in Arabidopsis,and 24 CmDEADs had collinearity relationships with 26 genes in cucumber.Gene structure analysis indicated that the average number of exons for the 29 CmDEADs was 10.38,with the number of exons ranging from 4-15.Conserved motif analysis revealed that all 29 genes contained the nine core motifs of the DEAD-box gene family.Transcriptome data analysis demonstrated that CmDEAD04,CmDEAD08,CmDEAD12,CmDEAD13,CmDEAD19 and CmDEAD25 maintained high expression levels at various development stages.Specifically,CmDEAD04 showed an upward trend in expression from 10-20 days after flowering and its expression began to decline 30 days after flowering in both datasets of GSE178084 and PRJNA286120,which suggested that CmDEAD04 may play a significant role in melon fruit development.A theoretical reference for genetic research on the melon genome and the DEAD-box family are provided in this study.
