摘要

Abstract

In order to optimize and improve the strawberry stem apex tissue culture seedling technology.taking'Benihoppe'strawberry as the test material and stem apex as the explant,based on the MS medium,the optimal developmental state of the collected stem apex,the optimal time for sodium hypochlorite disinfection,the optimal ratio of the propagation and rooting medium to the substrate for seedling growth were investigated,and the flowering and fruiting conditions of the tissue culture seedlings were observed.The results showed that the optimal developmental state for collecting the stem apex of'Benihoppe'strawberries was when the first leaf of the stolon was less than 1/2 unfolded and the second leaf was not fully unfolded.Disinfection was most suitable with alcohol treatment for 30 seconds and 2.5% sodium hypochlorite treatment for 10 minutes.The optimal propagation medium was MS+6-BA(0.5 mg/L).The optimal rooting medium was 1/2 MS+IBA(0.5 mg/L).The optimal substrate volume ratio for seedling growth was weathered sand∶peat soil∶garden soil=1∶1∶1.After the tissue culture seedlings were transplanted,the flowering and fruiting were normal.This plan could provide a reference for the factory-based seedling cultivation of strawberries.
