Pathogenic fungal identification and pathogenicity verification of leaf blight disease of ancient Platycladus orientalis in Beijing

Abstract
Leaf blight disease of ancient Platycladus orientalis was common in Beijing,severely affecting its survival and ornamental value.The pathogen was isolated from two leaf specimens of leaf blight disease collected from the Changling Tomb and the Summer Palace in Beijing.The pathogen was preliminarily identified by morphological observation and combined with internal transcribed spacer(ITS),glyceraldehyde-3-phosphate dehydrogenase(GAPDH)and translation elongation factor1-alpha(TEF1)multi-gene phylogenetic analysis,and the pathogenicity verification test was carried out.Fungal strains were identified as two species of Bipolaris,namely Bipolaris sorokiniana and Bipolaris setariae.Koch's postulates confirmed that both pathogenic fungi could cause leaf blight disease of P.orientalis.This study is the first to report leaf blight disease of P.orientalis caused by Bipolaris spp.,providing a basis for the diversity of pathogenic fungi causing leaf blight disease and disease management of ancient P.orientalis in Beijing.关键词
