Abstract

To study the ability of Pyemotes zhonghuajia to carry Beauveria bassiana spores as well as activity and virulence of the spores carried by P.zhonghuajia,and evaluate potential of the combination application,based on low pathogenicity of B.bassiana BbFz-91 against P.zhonghuajia,B.bassiana spore powder spraying method was used to observe spores carried on surface of P.zhonghuajia at 12,24,48 h post-treatment by scanning electron microscopy.Number and activity of spores were quantitatively measured by using hemocytometer and colony counting method,and virulence of spores carried by P.zhonghuajia against Monochamus alternatus larvae was determined.The results showed that B.bassiana spores could be carried on body surface of P.zhonghuajia.After 12 h treatment,an average of 1.54×105 spores were carried per P.zhonghuajia,spores carried and transmitted by 10 of P.zhonghuajia formed an average of 33 colonies on medium,and cumulative corrected mortality rate of M.alternatus larvae reached 80.65%10 d after treated with P.zhonghuajia spraying B.bassiana spore powder.Number of spores carried and number of colonies formed gradually decreased with treatment time increasing,and corrected mortality of larvae also gradually decreased.Spores carried by P.zhonghuajia after 12 h spraying spore powder treatment had the shortest lethal median time to larvae of M.alternatus and lethal median time was significantly prolonged after 72 h treatment.P.zhonghuajia had ability to actively spread spores of B.bassiana,and scattered spores had certain activity and toxicity.It was feasible that combinating P.zhonghuajia and B.bassiana to control M.alternatus in forest,and the results laid a theoretical foundation for further research on application technology in forest.
