摘要

Abstract

Objective To construct a nanoprobe that can precisely target malignant pleural mesothelioma(MPM)and evaluate its therapeutic effect and magnetic resonance imaging(MRI)chemical exchange saturation transfer(CEST)imaging performance.Methods Pemetrexed(Pem)was encapsulated in zeolite imidazolate framework material(ZIF-8)by surface adsorption method to prepare nano-drug ZIF-8@Pem.ZIF-8@Pem was characterized by transmission electron microscopy and dynamic light scattering,and its activation efficiency for MRI-CEST was detected.Zinc ion uptake experiment was used to clarify the cellular uptake,cytotoxicity experiment and JC-1 staining were used to explore its killing effect on tumor cells,and flow cytometry was used to detect the cell apoptosis.A subcuta-neous transplanted tumor mouse model of pleural mesothelioma was established and then injected with PBS,ZIF-8 and ZIF-8@Pem nanoparticles via the tail vein,respectively.The tumor size was measured,and its antitumor effect in vivo was evaluated by Caspase-3 staining.Results The prepared ZIF-8@Pem nanoprobe had a polyhedral structure with an average particle size of about 55 nm,which had good stability and CEST activation efficacy.Zinc ion uptake experiments showed that the nanomaterial was efficiently taken up by cells.Flow cytometry results showed that the early and late apoptosis of MSTO-211H cells was significantly increased after co-incubation with ZIF-8@Pem compared with that after co-incubation with ZIF-8(P<0.001).In vivo experiments showed that the tumor volume of nude mice was significantly lower after treated with ZIF-8@Pem than that after ZIF-8 treatment(P<0.001),and the area of Caspase-3 yellow-brown stained area was significantly elevated.Conclusion The nanomedicine ZIF-8@Pem is successfully prepared,and it can specifically activate CEST imaging and improve the target and the therapeutic efficacy,which provides a new idea for the diagnosis and treatment of pleural mesothelioma.
