Abstract

Objective To explore the regulatory effect of miR-21-3p on kallikrein-related peptidase 11(KLK11),and the regulatory effect and mechanism of miR-21-3p/KLK11 axis on the radiosensitivity of laryngeal squamous cell carcinoma(LSCC)cells.Methods①Human laryngeal carcinoma cell line AMC-HN-8 cells were divided into si-NC group,si-KLK11 group,oe-NC group,and oe-KLK11 group,and the cells were treated with different doses of X-ray irradiation(0,2 4,6,8 Gy)in each group,respectively.The cell proliferation was detected by CCK-8 and clonal formation assay.2The targeting binding relationship between miR-21-3p and KLK11 was analyzed by double luciferase reporter gene assay.AMC-HN-8 cells were divided into oe-NC group,oe-KLK11 group,oe-KLK11+mimics NC group and oe-KLK11+miR-21-3p group.The cells in each group were treated with X-ray irradiation with a dose of 6 Gy.The cell proliferation was detected by CCK-8 and clonogenesis assay,the cell apoptosis was detected by flow cytometry,the cell migration was detected by cell scratch assay,the cell invasion was detected by Transwell assay,the levels of miR-21-3p and KLK11 gene were detected by RT-qPCR,and the protein expression levels of KLK11,p53,Bcl-2,Bax,E-cadherin,Vimentin,N-cadherin,β-catenin and Wnt1 were detected by Western blot.Results ①Compared with si-NC group,the cell proliferation and the number of clone formation of AMC-HN-8 cells were increased after X-ray treatment at doses of 2,4,6,8 Gy in si-KLK11 group(P<0.05).Com-pared with oe-NC group,the cell proliferation and the number of clone formation of AMC-HN-8 cells in oe-KLK11 group were de-creased under X-ray treatment at doses of 0,2,4,6 8 Gy(P<0.05).2There was a targeted binding relationship between miR-21-3p and KLK11.Compared with oe-NC group,the levels of cell proliferation,migration and invasion were decreased in oe-KLK11 group(P<0.05),the apoptosis rate was increased(P<0.05),and the protein levels of Wnt1 and β-catenin were decreased(P<0.05).Com-pared with oe-KLK11+mimics NC group,the levels of cell proliferation,migration and invasion were increased in oe-KLK11+miR-21-3p group(P<0.05),the apoptosis rate was decreased(P<0.05),and the protein levels of Wnt1 and β-catenin were increased(P<0.05).Conclusion miR-21-3p targets KLK11 to promote the proliferation,migration,invasion and epithelial-mesenchymal transi-tion of LSCC cells irradiated by X-ray,and inhibits the apoptosis of LSCC cells irradiated by X-ray,which may be related to the activa-tion of Wnt/β-catenin signaling pathway.
