Effect of long noncoding RNA WDFY3-AS2 on proliferation,invasive metastasis and epithelial-mesenchymal transition in head and neck squamous cell carcinoma

Abstract
Objective To investigate the effects of the long noncoding RNA WDFY3-AS2 on the proliferation,invasion,metastasis and epithelial-mesenchymal transition(EMT)of head and neck squamous cell carcinoma(HNSCC)cells,and to explore the underlying molecular mechanisms.Methods In vitro experiments were conducted using the WSU-HN30 cell line,with WDFY3-AS2 knockdown(sh-WDFY3-AS2)and overexpression(OE-WDFY3-AS2)models.The experiment was divided into WT group(wild type,no treatment),sh-NC group(knockdown lncRNA WDFY3-AS2),sh-WDFY3-AS2 group(knockdown lncRNA WDFY3-AS2),OE-NC group(overexpression lncRNA WDFY3-AS2)and OE-WDFY3-AS2 group(overexpression lncRNA WDFY3-AS2).Cell proliferation was assessed using cell counting kit-8 and 5-ethynyl-2'-deoxyuridine(EdU)assays,while cell migration and invasion were evaluated through scratch wound healing and Transwell assays.The expression levels of E-cadherin,N-cadherin and Vimentin were measured by Western blot.Results Knockdown of WDFY3-AS2 significantly enhanced HNSCC cell viability and proliferation.The cell counts at 72 hours of sh-WDFY3-AS2 group and sh-NC group were 177.20±13.16 and 98.55±6.49,the proportion values of EdU-positive cells were(32.12±2.29)%and(18.77±0.73)%,percentage of wound healing area were(80.03±5.13)%and(59.36±3.26)%,number of invasion cells was 259.67±44.16 and 131.00±19.97,the relative expression levels of N-cadherin were 0.75±0.03 and 0.40±0.01,the relative expression levels of Vimentin were 1.01±0.02 and 0.86±0.03,the relative expression levels of E-cadherin were 0.51±0.01 and 0.71±0.02;the differences of above indexes were statistically significant(P<0.01,P<0.001).Overexpression of WDFY3-AS2 exhibited opposing trends.Additionally,there was a positive correlation between WDFY3-AS2 and BRAF expression(P<0.05).Knockdown or overexpression of WDFY3-AS2 substantially decreased or increased BRAF mRNA and protein levels,respectively.Conclusion WDFY3-AS2 inhibits HNSCC cell proliferation,invasion,metastasis,and EMT progression by regulating BRAF expression,it can serve as a potential therapeutic target for HNSCC,offering new strategies for clinical intervention.关键词
