Effects of ellagic acid on migration,pyroptosis and DNA damage of endometrial cancer cells

Abstract
Objective To investigate the effects of ellagic acid on the migration,pyroptosis,and DNA damage of endometrial cancer cells by inhibiting microRNA-331-3p(miR-331-3p).Methods AN3CA cells in the logarithmic growth phase were divided into the control group(normal culture,without any treatment),the experimental group(AN3CA cells treated with 20 μmol·L-1 ellagic acid),the mimics NC group(AN3CA cells transfected with mimics NC and then treated with 20 μmol·L-1 ellagic acid),and the miR-331-3p mimics group(AN3CA cells transfected with miR-331-3p mimics and then treated with 20 μmol·L-1 ellagic acid).Real-time fluorescent quantitative quantitative polymerase chain reaction was used to detect miR-331-3p expression levels,fluorescence in situ hybridization was used to detect miR-331-3p expression levels;5-ethynyl-2'-deoxyuridine was used to detect cell proliferation rate;cell scratch assay was used to detect cell migration rate;Western blot was used to detect the relative expression levels of NOD-like receptor thermal protein domain associated protein 3(NLRP3),gasdermin E(GSDME),histone family 2A variant(H2AX),phosphorylated histone H2AX(γ-H2AX),ataxia telangiectasia mutated(ATM)and phospho-ATM(p-ATM)proteins.Results The relative expression levels of miR-331-3p in the control and the experimental groups were 1.00±0.16 and 0.71±0.11,respectively;the relative fluorescence intensities of miR-331-3p were 1.00±0.13 and 0.58±0.06,respectively.The cell proliferation rates in the control group,the experimental group,the mimics NC group and the miR-331-3p mimics group were(84.23±6.54)%,(34.56±5.77)%,(32.39±5.12)%and(64.05±6.59)%,respectively;the cell migration rates were(71.33±6.32)%,(30.22±4.68)%,(32.17±4.52)%and(66.04±5.15)%,respectively;the relative expression levels of NLRP3 protein were 1.00±0.14,2.26±0.39,2.18±0.34 and 1.55±0.27,respectively;the relative expression levels of GSDME protein were 1.00±0.15,3.74±0.58,3.78±0.61 and 2.03±0.38,respectively;the ratios of γ-H2AX/H2AX protein were 1.00±0.12,2.98±0.41,3.02±0.45 and 1.96±0.35,respectively;the ratios of p-ATM/ATM protein were 1.00±0.16,2.35±0.39,2.39±0.40 and 1.63±0.28,respectively.The differences in the above indicators between the experimental group and the control group,and between the miR-331-3p mimics group and the mimics NC group,were statistically significant(P<0.05,P<0.01,P<0.001).Conclusion Ellagic acid may inhibit the proliferation and migration of cancer cells,induce pyroptosis and DNA damage by inhibiting miR-331-3p expression,thereby inhibiting the progression of endometrial cancer.关键词
