中度宫腔粘连患者子宫内膜组织单细胞转录组测序研究
Single-cell transcriptomic sequencing study of endometrial tissue in patients with moderate intrauterine adhesions

Abstract
Objective To analyze cellular differences between distinct tissues within the same environment and explore the pathogenesis of IUA by performing single-cell sequencing on both the adhesional endometrium and normal endometrium from the same intrauterine adhesion(IUA)patient.Methods One patient with moderate IUA who underwent hysteroscopic adhesiolysis at Huzhou Maternity & Child Health Care Hospital on December 20,2021,was enrolled in this study.Both adhesional endometrial tissue and normal endometrial tissue were collected from this patient during hysteroscopy and used as research specimens.The samples were immediately sent for examination and strictly processed in accordance with single-cell RNA sequencing protocols,including sequencing,data alignment,cell clustering,and differential gene expression analysis.Results A total of 21 284 single cells were obtained for subsequent data analysis,including 11 054 cells from the adhesion endometrium group and 10 230 cells from the normal endometrium group.Nine major cell types were identified:fibroblasts,epithelial cells,natural killer cells,T cells,ciliated epithelial cells,endothelial cells,macrophages,B cells,and mast cells.It was found that fibroblasts were increased in the adhesional endometrial tissue.Subpopulations of fibroblasts were associated with metabolic pathways related to energy metabolism and mitochondrial respiration,representing unique functional phenotypes in the context of adhesion.Conclusion The formation of intrauterine adhesions may be related to energy metabolism and mitochondrial respiration in endometrial tissue,with fibroblasts potentially playing a key role in the development of adhesions.关键词
