Abstract

Objective To explore the impact of Helicobacter pylori(Hp)infection on the structure and biological functions of gastric mucosal microbiota in patients with gastric ulcer from a metagenomic perspective.Methods Eight patients with gastric ulcers who underwent gastroscopy at the First People's Hospital of Xiaoshan District,Hangzhou from March to June,2024 were selected as the observation group.In addition,eight healthy individuals aged 30-55 years with normal gastroscopy results at the same hospital during the same period were selected as the control group.Metagenomic sequencing was used to analyze the composition of the microbial community and the metabolic functions of related genes in the gastric mucosa tissue of the two groups of research subjects.Results In the observation group with Hp-positive patients,the gastric mucosal microbiota was dominated by Hp,and the biodiversity was significantly lower than that of the control group(P<0.05).In addition,compared to the control group,13 differential functional pathways were identified in the observation group,mainly involving energy metabolism,nucleotide and nucleotide biosynthesis,vitamin and cofactor biosynthesis,nucleotide metabolism,and other biosynthetic pathways.The differential functions of mucosal microorganisms were all derived from Hp,including queuosine-transfer RNA modification pathways and pathways related to demethylna-phthoquinone/naphthoquinone synthesis.Conclusion Hp infection significantly alters the structure,diversity,and biological functions of the gastric microbiota,potentially leading to mucosal damage and ulcer formation.
