Characteristics of SPAD spatial distribution in Ginkgo biloba leaf layers and its relationship with nitrogen content

Abstract
[Objective]Accurate,rapid and non-destructive diagnosis of Ginkgo biloba nitrogen status is conducive to nitrogen fertilization regulation of Ginkgo biloba,which can effectively improve Ginkgo biloba growth and development,yield and quality.[Method]The three-year old big Buddha finger Ginkgo biloba seedlings were taken from Baima base as the object,and used SPAD chlorophyll meter to study the leaves of different levels of Ginkgo biloba seedlings at different periods in 2020.[Result]1)With the growth of Ginkgo biloba seedlings within a year,the nitrogen content in the leaves gradually decreased,while the nitrogen content accumulated in the plant body gradually increased,and the two nitrogen indexes showed a trend of gradual stabilization after June;2)There was a good correlation relationship between SPAD indexes and nitrogen nutritional indexes of Ginkgo biloba seedlings,but it was not stable,and was greatly affected by the leaves at different times of the year and at different height levels.The most stable relationship between SPAD values and nitrogen content was found in the middle leaves of Ginkgo biloba seedling leaves,which was not limited by the period,and the correlation coefficient of determination reached the maximum in June(R2=0.56,P<0.001);whereas it was more unstable with nitrogen accumulation,and the correlation coefficient of determination reached the maximum in September(R2=0.60,P<0.000 1);3)The correlation of the SPAD indexes with the leaf nitrogen content and nitrogen accumulation,the correlation between SPAD and leaf nitrogen content was more stable,and thus more suitable as an effective technical means for nitrogen content estimation in Ginkgo biloba seedlings;4)Three single-period models and four full-period models were established based on the relationship between L2 and LNC,and all of them could achieve better nitrogen content estimation effects through comparison and verification.Among them,the single-period models,the April model(R2=0.53,RMSE=0.18)and the June to September model(R2=0.37,RMSE=0.28)are more reliable;the binary quadratic regression model of the full-period model has the highest accuracy of model measurement(R2=0.62,RMSE=0.26).[Conclusion]The SPAD chlorophyll meter method can be used as a simple method for evaluating the nitrogen nutritional status of Ginkgo biloba seedlings,and can provide a simple and feasible technical way to monitor nitrogen fertilisation in forest trees.关键词
