Research Progress on Comprehensive Morphological and Microscopic Authentication of Paris polyphylla and ITS2 Sequence Analysis

Abstract
Based on identification techniques including morphological characteristics,microscopic features,physicochemical properties and DNA sequences,this paper reviews and analyzes the differences between genuine Paris polyphylla and its adulterants to provide references for authentication.The varieties of genuine P.polyphylla and its adulterants were summarized based on literature,including differences in original plant morphology and microscopic characteristics.The ITS2 sequences of P.polyphylla and its adulterants were downloaded from public databases,standardized based on Hidden Markov Model principles,and subjected to sequence alignment,secondary structure prediction and phylogenetic tree cluster analysis.The main adulterants of genuine P.polyphylla include P.fargesii,P.latifolia,P.thibetica and P.forrestii,while common counterfeits are Polygonum bistorta,Polygonum viviparum and Typhonium giganteum.Significant differences exist between genuine and adulterant materials in plant morphology(stem,leaf,flower,fruit and seed)and microscopic identification,while crude drug characteristics and microstructures show minor differences.In DNA sequence identification,the ITS2 sequences show minor differences between genuine and adulterant materials but significant differences from counterfeits.Genuine and adulterant materials can be effectively distinguished by plant morphology,while ITS2 sequences and their secondary structures can rapidly and accurately identify counterfeits when only crude drugs are available.This study provides references for clinical safety and quality evaluation of P.polyphylla,and may also serve as reference for authentication of other medicinal materials.关键词
