山茶WRKY基因家族鉴定及其在花色形成中的表达分析
Identification and expression analysis of WRKY gene family in flower color formation in Camellia japonica

Abstract
[Objective]The WRKY gene family plays an important role in the regulation of plant growth and development.Camellia japonica is an important ornamental plant with rich flower colors.It is of great significance to identify and analyze the WRKY gene family members in Camellia japonica petals with different colors.[Method]Based on the whole genome data of Camellia japonica,the WRKY gene family was identified and analyzed by bioinformatics methods.Based on the transcriptome data related to camellia flower color,the expression pattern of CjWRKYs in different colored camellia petals was analyzed.[Result]The results indicated that 94 CjWRKY genes were identified in the whole genome and named CjWRKY1-CjWRKY94.The phylogenetic tree divided the strains into three groups(Ⅰ,Ⅱ and Ⅲ),and group Ⅱ was further divided into five subgroups(Ⅱ-a,Ⅱ-b,Ⅱ-c,Ⅱ-d and Ⅱ-e).Group I members contained two WRKY domains,while group Ⅱ and Ⅲ members contained only one WRKY domain.A total of 10 conserved motifs and 5 conserved domains were identified.The CjWRKYs members in the same subgroup had similar conserved motifs,conserved domains and gene structures.The CjWRKYs promoter region contained 38 elements for light response,stress response and hormone response,including MYB binding site for regulating flavonoid biosynthesis.94 CjWRKYs were unevenly distributed on 15 chromosomes,and there were 13 pairs of tandem duplication events.Collinearity analysis showed that there were 72 duplication events in 94 CjWRKYs genes,and the Ka/Ks value of the duplication genes was less than 1.There were 116 collinear gene pairs with Arabidopsis WRKY gene family.Among them,32 CjWRKYs were up-regulated and 6 CjWRKYs were down-regulated with the deepening of flower color of Camellia japonica,which suggested that CjWRKYs played an important role in the regulation of anthocyanin synthesis.It is speculated that CjWRKY48,CjWRKY53 and CjWRKY66 positively regulate anthocyanin synthesis by interacting with MYB transcription factors,thereby affecting flower color of Camellia japonica.[Conclusion]94 CjWRKYs genes were identified in the whole Camellia japonica genome,and most CjWRKYs are involved in anthocyanin synthesis and play an important role in color formation of Camellia japonica.关键词
