Abstract

In order to investigate the causative agent of a suspected CPV infection case and elucidate the genetic evolution of circulating strain,this study employed a comprehensive approach integrating clinical symptom evaluation,histopathological examination,and molecular diagnostics,and the VP2 gene of the identified CPV strain was then amplified for subsequent sequencing,followed by BLAST analysis,amino acid mutation analysis and phylogenetic evolution analysis.The results of the colloidal gold immunochromatographic assay and CPV-specific PCR targeting intestinal contents confirmed were all positive,which providing conclusive evidence of lethal CPV infection,and named the infected strain as CPV-HN.Then,the VP2 gene of CPV-HN was amplified with a 1755 bp product and sequenced.The nucleotide sequence analysis demonstrated the VP2 gene of CPV-HN shared 100%identity with the CPV-HN1617 strain,while sharing 97.7-99.9%identity with other reference strains.Moreover,the amino acid mutation analysis revealed the VP2 protein of CPV-HN contains a characteristic glutamic acid at position 426,and the phylogenetic analysis showed that the CPV-HN was clustered within the CPV-2c genotype clade,which provides reference for understanding the prevalence of CPV in this region.
