Abstract

In the development of new anti-coccidiosis vaccines and drugs,in vitro culture technology can serve as an important technical platform for studying the developmental mechanism of coccidiosis and high-throughput screening of anti coccidiosis drugs.In order to find a cell culture system suitable for the invasion and development of coccidiosis,the isolation and cultivation techniques of chicken embryo intestinal epithelial cells were established and optimized.Using transgenic Eimeria fetida expressing yellow fluorescent protein as a model,its spores were inoculated into chicken embryo intestinal epithelial cells,and the developmental process at each stage was observed to evaluate the efficiency of chicken embryo intestinal epithelial cells as an in vitro culture system for Eimeria fetida.The results showed that chicken embryo intestinal epithelial cells were isolated using 180 mesh and 500 mesh cell sieves,and the cultured cells exhibited typical epithelial cell characteristics.After about 66 hours of invasion into chicken embryo intestinal epithelial cells,the spores of genetically modified Eimeria fetida develop into the first generation mature schizonts,and release the first generation schizonts around 78 hours.Transgenic Eimeria spores can complete the development of trophozoites and schizonts in chicken embryo intestinal epithelial cells.On the 5th to 6th day of culture,we observed a small amount of large gametes and zygote morphology,indicating that Eimeria can engage in sexual reproduction and development in intestinal epithelial cells.This study provides excellent technical support for research on the development of Eimeria,and also has reference significance for the study of other intestinal pathogens.
