黄连解毒汤调控NLRP3炎症小体改善SHR血管功能的研究
Study on the Regulation of NLRP3 Inflammasome by Huanglian Jiedu Decoction to Improve Vascular Function

Abstract
Objective:To investigate the effects of Huanglian Jiedu Decoction on the expression of NOD-like receptor protein 3(NLRP3)inflammasome in the aortic tissue of spontaneously hypertensive rats(SHR).Methods:SHRs were randomly divided into a model group,a captopril group,and low,medium,and high-dose Huanglian Jiedu Decoction groups.Wistar-Kyoto(WKY)rats served as the blank control group.Following intragastric administration of the respective treatments,pathological changes in the thoracic aorta were examined using HE and Masson staining.Serum levels of interleukin-1β(IL-1β),interleukin-18(IL-18),endothelial nitric oxide synthase(eNOS),and endothelin-1(ET-1)were assessed by ELISA.Expression of caspase-1,NLRP3,gasdermin D(GSDMD),and apoptosis-associated speck-like protein(ASC)mRNA in the aorta was measured by qRT-PCR.Caspase-1 protein expression in the aorta was analyzed by immunohistochemistry.Results:Pathological staining indicated improvement in vascular pathology in both the Huanglian Jiedu Decoction and captopril groups compared to the model group.ELISA results showed that serum levels of IL-18,IL-1β,and ET-1 were lower,and eNOS levels higher,in the captopril group compared to the model group(P<0.05).In the medium-dose Huanglian Jiedu group,IL-1β and ET-1 levels decreased(P<0.05),and in the high-dose Huanglian Jiedu group,IL-18,IL-1β,and ET-1 levels decreased,while eNOS levels increased(P<0.05).qRT-PCR results showed that ASC,caspase-1,GSDMD,and NLRP3 expression levels were lower in the captopril group compared to the model group(P<0.05).In the medium-dose Huanglian Jiedu group,caspase-1,GSDMD,and NLRP3 expression levels decreased(P<0.05),while in the high-dose group,ASC,caspase-1,GSDMD,and NLRP3 expression levels also decreased(P<0.05).Immunohistochemistry showed a decrease in caspase-1 expression in both the Huanglian Jiedu Decoction and captopril groups compared to the model group(P<0.05).Conclusion:Huanglian Jiedu Decoction may protect vascular endothelial function in SHR by regulating the expression of molecules associated with the NLRP3 inflammasome in the thoracic aorta,thereby reducing the expression of inflammatory factors.关键词
