Abstract
Objective:To explore the effect of glyceraldehyde-3-phosphate dehydrogenase(GAPDH)expression on rats with spleen deficiency and dampness obstruction type simple obesity,and to investigate the role of GAPDH protein in the pathogenesis of this model through acupuncture intervention.Methods:Forty healthy SD rats were randomly divided into four groups:control group,model group,acupuncture group,and acupuncture control group,with 10 rats in each group.After successful modeling,pathological changes in the liver,spleen,and adipose tissue of rats in different groups were observed using HE staining under a light microscope.The levels of leptin and chemerin in rat serum were measured using double antibody sandwich ELISA.GAPDH mRNA expression was detected by quantitative PCR,and GAPDH protein expression was measured by Western blot.Results:The experimental results showed that the average body weight of rats in the model group was 20%higher than that of the control group,and the body weight of rats in the model group increased significantly compared to the control group(P<0.01).Pathological changes in the liver,spleen,and adipose tissues of rats were observed after HE staining.Compared to the control group,the model group exhibited varying degrees of pathological changes.The serum leptin levels were higher in the model group compared to the control group(P<0.05),and the acupuncture group had lower levels than the model group(P<0.05),while the acupuncture control group showed no significant difference(P>0.05).The serum chemerin levels were higher in the model group compared to the control group(P<0.05),and the acupuncture group showed a decrease compared to the model group(P<0.05),with no significant difference in the acupuncture control group(P>0.05).PCR results showed that GAPDH mRNA expression in the spleen tissue of rats in the model group was higher compared to the control group(P<0.01).After acupuncture treatment,the expression of GAPDH mRNA in the acupuncture group was significantly lower than in the model group(P<0.01),and there was no significant difference between the acupuncture control group and the model group(P>0.05).Western blot results showed that the expression of GAPDH protein in the spleen tissue of rats in the model group was significantly higher than in the control group(P<0.01).After acupuncture treatment,the GAPDH protein levels in the acupuncture group and acupuncture control group were significantly reduced compared to the model group(P<0.05).Conclusion:Glyceraldehyde-3-phosphate dehydrogenase(GAPDH)is a promising specific protein for spleen deficiency and dampness obstruction syndrome,providing a basis for early diagnosis of this syndrome.关键词
