胁腹宁颗粒制备工艺优化及薄层鉴别和含量测定研究
Optimization of Preparation Process and Establishment of Quality Standard for Xiefuning Granule

Abstract
Objective:To optimize of the preparation process and establishment of TLC and content determination method for Xifun-ing granule.Methods:Dry extract rate,aloe-emodin,chrysophanol and emodin monomethyl ether total content were examined as in-dicators and water addition,extraction time and times were examined as factors,and orthogonal test was used to optimize the best extraction process;the granula molding process was determined using formation rate as the index of investigation and type and dos-age of accessories,concentration and dosage of wetting agent and drying temperature as the factors of investigation;the grouped herbs were identified by TLC;the content determination of aloe-emodin,chrysophanol,emodin monomethyl ether was determined by HPLC.Results:The optimal preparation process was soaking for 2 hours,extracted three times,water addition of 12,10,10 times,1 hour each time,drug-assisted ratio was 10:7,95%ethanol volume fraction,35%wetting agent dosage,screening,drying at 60℃,packaging;TLC of Rhei Radix Et Rhizoma,Corydalis Rhizoma,Desmodii Styracifolii Herba,Gentianae Radix Et Rhizoma,and Glycyrrhizae Radix Et Rhizoma were established and the spectrum were well spotted and negative without interference;the three components showed good linear relationships within their own ranges(r≥0.9990),whose average recoveries were 96.60%~102.19%with the RSDs of 0.29%~0.40%.The total anthraquinone content of each bag of granules should not be less than 2.6 mg.Conclusion:The preparation process and TLC and content determination method of Xiefuning granule are stable and feasible,pro-viding a reference for the development of this preparation.关键词
