盐酸青藤碱对急性放射性肺损伤的影响及作用机制研究
Effects and mechanism of sinomenine hydrochloride on acute radiation-induced lung injury

Abstract
Objective To investigate the effects and underlying mechanism of quinamine hydrochloride on acute radiation-induced pulmonary injury.Methods A total of 25 male C57BL/6J mice aged 8 weeks were randomly assigned to 5 groups(5 mice per group):the control group(A),the radiation-only group(B),the low-dose quinamine hydrochloride group(C),the high-dose quinamine hydrochloride group(D),and the dexamethasone group(E).Except for the blank control group,the chests of the remaining four groups of mice were subjected to a single dose of 18 Gy of 6 MV X-rays.Starting from the second day after X-ray irradiation,the blank control group and the irradiation-only group were given normal saline,while the low-and high-dose sinomenine hydrochloride groups were given 40 and 80 mg/kg of sinomenine hydrochloride,respectively,and the dexam-ethasone group was given 2 mg/kg of dexamethasone.The five groups of mice were administered the above treatments by gavage continuously for 4 weeks,with a gavage dose of 10 μL/(god).After 4 weeks of gavage,the mice were sacrificed,and the weight of the lung tissues was measured,and the lung coefficient was calculated.The expression levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in the serum were detected by ELISA.Histopathological changes in lung tissue were examined by hematoxylin and eosin(H&E)staining,and collagen deposition was assessed by using Masson's trichrome staining.Western blotting was performed to analyze the expression of epithelial-mesenchymal transition(EMT)markers(E-cadherin,N-cadherin,and Vimentin).Immunohistochemistry was used to evaluate the expression of proteins involved in the TGFβ1/Smad signaling pathway.Results Compared to the control group,the radiation-only group exhibited a significant increase in lung coefficient,a-long with alveolar damage,extensive inflammatory cell infiltration,and substantial collagen deposition.Additionally,levels of TNF-α and IL-1β were elevated,and the TGFβ1/Smad signaling pathway,along with the EMT process,was activated.In con-trast,the quinamine hydrochloride and dexamethasone treatment groups showed partial alleviation of lung injury,with reduced TNF-α and IL-1β levels,and effective inhibition of both the TGFβ1/Smad signaling pathway and the EMT process.Conclusion Sinomenine hydrochloride may alleviate acute radiation-induced lung injury in mice by inhibiting the TGFβ1/Smad signaling pathway and the epithelial-mesenchymal transition process.关键词
