基于PI3K/AKT信号通路探讨白芍总苷胶囊对非小细胞肺癌增殖、迁移、侵袭和凋亡的影响
Effects of total glucosides of White Paeony capsules on proliferation,migration,invasion and apoptosis of non-small cell lung cancer based on PI3K/AKT signaling pathway

Abstract
Objective:To investigate the effects of total glucosides of White Paeony capsules(TGP)on proliferation,migra-tion,invasion and apoptosis of non-small cell lung cancer based on the PI3K/AKT signaling pathway,and explore its mechanism.Methods:CCK-8 was used to detect the effects of different concentrations of TGP on A549 and H1975 cell viability.On the basis of CCK-8 results,A549 and H1975 cells were classified into five groups:the control group(no drugs),the TGP low dose group(TGP-L,1 g/L),the TGP high dose group(TGP-H,4 g/L),the LY294002 group(PI3K pathway inhibitor,10 nmol/L),the TGP high dose+LY294002 group(4 g/L+10 nmol/L).Cloning test was used to detect cell proliferation.Scratch test was used to detect cell migration.Transwell test was used to detect cell invasion.Flow cytometry was used to detect apoptosis.The expres-sions of EMT-related proteins(E-cadherin and N-cadherin),apoptosis related proteins(Bax,Bcl-2 and Cleaved caspase-3)and PI3K/AKT pathway related proteins(p-PI3K/PI3K and p-AKT/AKT)in A549 and H1975 cells were detected by Western blot.Results:Compared with the control group,the proliferation ability,migration ability and invasion ability of lung cancer A549 and H1975 cells in TGP low dose group,LY294002 group,TGP high dose group and TGP high dose+LY294002 group were de-creased,while the cell apoptosis rate was increased.The protein expression of N-cadherin,Bcl-2,p-PI3K/PI3K and p-AKT/AKT was decreased,while the protein expression of E-cadherin,Bax and Cleaved caspase-3 was increased.And the above indicators were better regulated in the TGP high dose group than in the TGP low dose group(all P<0.05);compared with the high dose TGP group or LY294002 group,the proliferation ability,migration ability and invasion ability of lung cancer A549 and H1975 cells in TGP high dose+LY294002 group were decreased,while the cell apoptosis rate was increased.The protein expression of N-cadherin,Bcl-2,p-PI3K/PI3K and p-AKT/AKT was decreased,while the protein expression of E-cadherin,Bax and Cleaved caspase-3 was increased(all P<0.05).Conclusion:TGP can inhibit the proliferation,migration and invasion of A549 and H1975 cells and promote their apoptosis,which may be related to the inhibition of the PI3K/AKT signaling pathway.关键词
