摘要

Abstract

Objective:To explore the effect and possible molecular mechanism of Biejiajian Pill(BJJP)-containing serum on the malignant biological behavior of pancreatic cancer AsPC-1 cells.Methods:A total of 72 healthy Sprague-Dawley(SD)rats were used to prepare normal rat serum and low-(0.55 g/kg),medium-(1.1 g/kg),and high-(2.2 g/kg)doses of BJJP-containing serum.Pancreatic cancer AsPC-1 cells in the logarithmic growth phase were divided into the blank control group(BC),normal rat serum treatment group(NC)and low-,medium-,and high-doses of BJJP-containing serum treatment groups(BJJP-L,BJJP-M,and BJJP-H);these cells in each group were treated with 10%corresponding serum for 48 h before subsequent experiments were performed.H&E staining was used to assess the effect of BJJP on the histological changes of liver,kidney,and heart of healthy SD rats.The proliferation ability of AsPC-1 cells was detected by CCK-8 assay;the colony formation ability of AsPC-1 cells was detected by colony formation assay;the apoptosis of AsPC-1 cells was detected by TUNEL assay;the migration of AsPC-1 cells was detected by scratch assay;the migration and invasion of AsPC-1 cells were detected by Transwell assay;the ex-pression levels of the proliferation-related proteins(Ki-67 and PCNA),apoptosis-related proteins(Caspase-3,Caspase-9,Bax,Bcl-xl,and Bcl-2),epithelial-mesenchymal transition(EMT)-related proteins(E-cadherin,N-cadherin,and Vimentin)and PI3K/AKT signaling pathway-related proteins(p-PI3K/PI3K and p-AKT/AKT)of AsPC-1 cells were detected by Western blot;the fluorescence intensity of EMT-related proteins(E-cadherin,N-cadherin,and Vimentin)of AsPC-1 cells were detected by immunofluorescence.UHPLC-MS/MS was used to detect the chemical components of BJJP-containing serum.Results:H&E staining showed that low-,medium-,and high-doses of BJJP had no obvious toxic on the main organs of healthy SD rats.Com-pared to the NC group,the OD value,colony formation number,scratch healing rate,migration number,invasion number,the protein expression levels of Ki-67,PCNA,Bcl-xl,Bcl-2,N-cadherin,Vimentin,p-PI3K/PI3K,and p-AKT/AKT,and the flu-orescence intensity of N-cadherin and Vimentin were significantly reduced;TUNEL apoptosis rate,the protein expression levels of Caspase 3,Caspase 9,Bax,and E-cadherin,and the fluorescence intensity of E-cadherin were significantly increased in the low-,medium-,and high-dose BJJP-containing serum groups(P<0.05).UHPLC-MS/MS results showed that a total of 67 blood prototype components in BJJP-containing serum,including 30 active ingredients such as homoarbutin,quercetin,and atrac-tylodin.Conclusion:BJJP-containing serum inhibits the malignant biological behavior of pancreatic cancer AsPC-1 cells,and its mechanism may be related to inhibiting the PI3K/AKT signaling pathway.
