Abstract

Objective:To investigate the expression of phosphoenolpyruvate carboxykinase 1(PCK1)in cholangiocarcinoma cells,its effects on cell proliferation and migration,and the underlying mechanisms through bioinformatics analysis.Methods:Dif-ferentially expressed genes in cholangiocarcinoma were identified using mRNA sequencing data from the GEO database.A protein-protein interaction(PPI)network was constructed using STRING and Cytoscape,and key regulatory networks and genes were identified.The expression levels and pathological characteristics of key genes were validated using the TCGA and HPA databases.KEGG and GSEA enrichment analyses were performed to explore the functions of differentially expressed genes and PCK1-associ-ated pathways.RBE cholangiocarcinoma cells were transfected with lentivirus to overexpress PCK1.Transfection efficiency was confirmed by RT-qPCR and Western blot.Experimental groups included a negative control(RBE group),an empty vector con-trol(RBE-NC group),an overexpression group(RBE-OE group),and a PPARγ inhibitor group(RBE-OE+GW9662 group).Cell proliferation was assessed by colony formation and CCK8 assays,while migration was evaluated using wound healing and Transwell assays.Western blot was employed to analyze pathway protein expression.Results:Bioinformatics analysis revealed PCK1 as a key regulatory gene in cholangiocarcinoma,with low expression in tumor tissues and potential involvement in the PPAR signaling pathway.Cellular experiments showed that PCK1 mRNA and protein levels were significantly increased in RBE cells after transfection.Compared to the negative control,PCK1 overexpression inhibited RBE cell proliferation and migration,in-creased PPARγ and PTEN protein levels,and decreased p-mTOR/mTOR levels.These effects were reversed in the inhibitor group.Conclusion:PCK1 may act as a key regulatory gene in cholangiocarcinoma and potentially suppresses cell proliferation and migration through the activation of the PPARγ/PTEN/mTOR signaling pathway.
