Role of ferroptosis in chronic wounds
摘要
Abstract
Ferroptosis,a novel iron-dependent form of regulated cell death,plays a pivotal role in the impaired healing of chronic wounds by mediating lipid peroxidation,iron dysregulation,and immune imbalance.In diabetic foot ulcers(DFU),pressure injuries(PI),and chronic venous leg ulcers(CVLU),ferroptosis is activated through distinct yet overlapping mechanisms involving iron metabolism,glutathione peroxidase 4(GPX4)suppression,lipid metabolic alterations,and immune microenvironment remodeling.Several therapeutic approaches targeting ferroptosis—such as iron chelators,antioxidants,immune modulators,and biomaterial-based delivery systems—have demonstrated wound-healing benefits in animal models.Notably,platelet-rich plasma(PRP)has shown clinical promise in promoting ulcer closure and reducing infection rates.Ferroptosis serves as a key pathological amplifier linking inflammation and tissue degradation,and offers tangible therapeutic potential.Integrating ferroptosis-related biomarkers(e.g.,GPX4,ferritin,malondialdehyde)with immune profiling and wound subtype analysis may provide a framework for precision treatment of refractory chronic wounds.关键词
