产细菌素肠球菌筛选鉴定及其对耐万古霉素菌跨膜抑菌效应的研究
Screening and identification of bacteriocin-producing Enterococcus and their trans-membrane inhibitory effects on vancomycin-resistant strains

Abstract
Bacteriocin is an antimicrobial peptide with efficient inhibitory effects on related species and is considered as one of the effective antibacterial alternatives for antibiotics.In this study,Enterococcus was specifically extracted from clams(Meretrix meretrix)farmed in Jiangsu Province,and non-invasive vancomycin-resistant E.faecium(VREfm)and Staphylococcus aureus(VRSA)were used as indicator strains to screen for enterococcal bacteriocins with the ability to inhibit vancomycin-resistant pathogens.The results showed that among the 36 strains of Enterococcus extracted from clams,two strains showed inhibitory effects on VREfm,which were identified as E.durans and E.hirae,respectively,one strain showed inhibitory effects against both VREfm and VRSA,which was identified as E.faecalis.This isolate produced inhibitory substances with typical characteristics of bacteriocins,such as high thermal stability,tolerance to low pH and easy decomposition by protease.In addition,the treatment of VREfm using crude bacteriocin produced by E.faecalis revealed that the inhibition of bacteria by E.faecalis showed an inhibition concentration-dependant feature,with the increasing inhibition concentration,the protein concentration,conductivity,nucleic acid content and alkaline phosphatase activity of the bacterial solution increased significantly(P<0.05);the microscopic results from fluorescence,transmission and scanning microscopy showed that E.faecalis had a disruptive effect on the cell wall structure of bacteria,forming a pore-like structure,causing cytoplasmic leakage and thus causing the trans-membrane inhibitory effects on the bacteria.关键词
