罗伊氏菌素的大肠杆菌抗菌活性及其分子机制研究
Study on antibacterial bioactivity and molecular mechanism of reuterin against Escherichia coli K-12

Abstract
Reuterin is active toward various enteropathogens,yeasts,fungi,protozoa and viruses,but the detailed mechanism of reuterin has not yet been fully elucidated.For this purpose,Lactobacillus reuteri strain ATCC 55730,a reuterin-producing bacterium,was used for bacterial fermentation.It was induced by addition of glycerol at the concentration of 200 mmol·L-1,and the concentration of reuterin produced by the strain was determined as 26.13 mmol·L-1.Additionally,the prepared reuterin showed good stability under three temperature conditions including-20℃,room temperature of 25℃ and 37℃.The antibacterial bioactivity against 8 selected standard pathogenic bacterial strains,including five subtypes of Clostridium perfringens,Salmonella sp.LT2,clinical isolates of Candida albicans(CA)and Escherichia coli K-88,was measured.The results showed that reuterin obviously inhibited the bacterial growth of the tested pathogenic strains.Based on the qualitative analysis of the minimum inhibitory concentration(MIC),the values for five subtypes of C.perfringens,Salmonella sp.LT2,C.albicans and E.coli K-88 were determined as 0.52-1.83 mmol·L1,1.12 mmol·L-1,1.67 mmol·L-1 and 0.79 mmol·L-1,respectively.A total of 60 resistant genes and 69 sensitive genes of reuterin against E.coli K-12 were screened from the Keio library.Among them,two genes cyaA and sdhB were identified as resistant,while three genes including mnmE,usg and rbfA were identified as sensitive.To further explore the molecular targets,a high-throughput genome-wide screening procedure was used to obtain the genes related to the antibacterial performance of reuterin.In addition,functional analysis of the target genes was carried out to obtain the functional clusters of genes for the antibacterial bioactivity of reuterin.Results showed that two targets,Δcya A and ΔyheS,were selected as potential core genetic targets.And the genetic interaction analysis between the two core genes was further analyzed.It showed that the biological functions of ΔcyaA and ΔyheS genes were mainly related to the formation of channel proteins in the cell membrane,and the drug efflux pumps,respectively.Thus,it is speculated that the two molecular pathways related to the two core genes are the molecular targets for the antibacterial activity of reuterin against Escherichia coli K-12.The results can provide a scientific reference for systematically revealing antibacterial mechanism of reuterin and promoting its practical application.关键词
