解淀粉芽孢杆菌FS6绿色荧光蛋白标记菌株的获得及其生物学特性
Acquisition of a Green Fluorescent Protein-labeled Strain of Bacillus amyloliquefaciens FS6 and Its Biological Characteristics

Abstract
Plasmid pHT01 with green fluorescent protein gene(gfp)was transformed into FS6 strain by electroporation,and transformants carrying green fluorescent markers and stable heredity were ob-tained.Colony morphology,growth characteristics and antifungal activity between transformants FS6-GFP and wild strain were studied.PCR amplification and fluorescence observation showed that the green fluorescent marker gene was successfully transformed into the biocontrol strain FS6,and the genetic stability was still 96.8%after 50 generations of subculture.There was no significant differ-ence between FS6-GFP transformant with green fluorescent marker and FS6 wild strain in biological characteristics and antibacterial activity against seven main pathogens of Panax ginseng,and the an-tifungal rate could range from 63.1%to 82.3%.Bacillus amyloliquefaciens FS6 has obvious inhibi-tory effect on the main pathogens of ginseng root rot disease and other ginseng diseases.Strain FS6 was fluorescently labeled to explore its ecological adaptability and colonization rules in ginseng plants.This study provides a foundation for further research on the colonization characteristics and biocontrol mechanism of FS6 strain in ginseng.关键词
