Abstract

To investigate the changes in the chemical composition and antioxidant activity of Glycyr-rhizae Radix et Rhizoma before and after instantaneous high-temperature sterilization,and to evalu-ate the application of instantaneous high-temperature sterilization technology in Glycyrrhizae Radix et Rhizoma,high-performance liquid chromatography(HPLC)was used to determine the contents of 8 monomers in Glycyrrhizae Radix et Rhizoma before and after sterilization.A fingerprint was estab-lished,and the similarity evaluation system for traditional Chinese medicine(TCM)chromatographic fingerprints was used to identify common peaks.Similarity analysis(SA)and cluster analysis(CA)were performed on the peak areas of the common peaks.Liquid chromatography-mass spectrometry(LC-MS)was employed to confirm the structures of the common peaks.1,1-diphenyl-2-picrylhydra-zyl(DPPH)method was used to determine the antioxidant activities of Glycyrrhizae Radix et Rhizoma samples and 8 monomers.The reaction rate of Glycyrrhizae Radix et Rhizoma samples with excessive DPPH was determined by HPLC.Grey relational analysis(GRA)and partial least squares regression analysis(PLS)were used for spectral efficiency analysis.The results showed that instantaneous high-temperature sterilization had obvious killing effect on the microorganisms in Glycyrrhizae Radix et Rhizoma,and little effect on the contents of 8 monomers and DPPH antioxidant activity after steriliza-tion.The similarity between Glycyrrhizae Radix et Rhizoma fingerprint before and after sterilization was greater than 0.990.Cluster analysis results also showed that there was no difference between the samples before and after instantaneous high-temperature sterilization.The structure of 24 common peaks was confirmed by LC-MS.Peaks 1-12 were flavonoid glycosides and aglycones regions,peaks 13-17 were triterpenoid saponins regions,and peaks 18-24 were flavonoid aglycones regions.The fingerprint showed obvious characteristics of compound aggregation.The reaction rate of glabridin,glycyrrhizin chalcone A,and glicoricone with excessive DPPH was more than 50%,indicating that the free radical scavenging reaction rate was faster than other substances.The spectrum effect rela-tionship analysis showed that the correlation coefficients(r)between the 24 common peaks and the antioxidant activity were greater than 0.500,among which the correlation coefficients with the anti-oxidant activity were greater for glabridin,glycyrrhizin chalcone A,liquiritigenin,liquiritin,licori-cidin,glicoricone and glycyrol(r>0.730).In conclusion,instantaneous high-temperature steriliza-tion has a significant killing effect on microorganisms in Glycyrrhizae Radix et Rhizoma,and it does not significantly affect its chemical composition content and antioxidant activity,providing a basis for the application of instantaneous high-temperature sterilization in Glycyrrhizae Radix et Rhi-zoma.
