Lipid-lowering and Antioxidant Activities of Phlorizin on Caenorhab-ditis elegans

Abstract
We investigated the effects of phlorizin on the antioxidant and lipid-lowering activities of Caenorhabditis elegans by using phlorizin obtained from the leaves of Lithocarpus litseifolius(Hance)Chun in the laboratory.The effects of phlorizin on longevity,motility,stress resistance,antioxidant enzyme activity,lipofuscin accumulation,triglyceride and cholesterol content determination,and oil red O staining of C.elegans were determined and the mechanism of action was elaborated by using fluorescence quantitative PCR to reveal the anti-aging and lipid-lowering effects of phlorizin on C.el-egans.The results show the lifespan of nematodes treated with 50,100 and 200 μmol/L concentra-tions was improved under normal conditions,heat stress conditions and oxidative stress conditions,and was accompanied by an increase in motility.Phlorizin also increased the activities of SOD and CAT antioxidant enzymes and decreased the accumulation of MDA and lipofuscin.Triglyceride and cholesterol contents in nematodes were reduced after feeding phlorizin,while oil red O staining re-sulted in fewer red areas and smaller lipid droplet sizes.Phlorizin significantly up-regulated the ex-pression of daf-16,nhr-49,aak-2,acs-2 and atgl-1 genes,and significantly down-regulated the ex-pression of daf-2,fat-5,fat-6 and fat-7 genes.It indicates that phlorizin may reduce lipid accumu-lation through SREBP signalling pathway,nhr-49-mediated nuclear hormone signalling pathway,in-sulin signalling pathway,as well as AMPKsignalling pathway to enhance the motility and longevity of C.elegans.关键词
