LncRNA PCAT1通过上调乳酸脱氢酶A促进胰腺癌细胞增殖和迁移的研究
LncRNA PCAT1 facilitates the proliferation and migration of pancreatic cancer cells by up-regulating the expression of LDHA
摘要
Abstract
The aim of this study was to reveal the molecular mechanism of lncRNA PCAT1 contributing to cell prolif-eration and migration in pancreatic cancer cells.The reverse transcription-quantitative real-time polymerase chain reaction(RT-qPCR)and Western blotting analyses were performed to measure the expression of the indicated genes.The RNA pull-down assay combined with Western blotting validation,RNA immunoprecipitation(RIP)-qPCR assay,protein stabil-ity assay,glucose uptake assay,and lactate production assay were conducted to investigate the molecular mechanism by which PCAT1 contributed to the proliferation and migration in pancreatic cancer cells.The results demonstrated that PCAT1 upregulated the expression of LDHA(lactate dehydrogenase A)in pancreatic cancer cells.PCAT1 may interact with LDHA protein,but had no effect on its stability and degradation in pancreatic cancer cells.Furthermore,PCAT1 promoted glucose uptake and lactate secretion by up-regulating the expression of LDHA,thereby promoted cell prolifera-tion and migration.In summary,our results suggested that PCAT1 promoted aerobic glycolysis by up-regulating the ex-pression of LDHA and interacting with LDHA protein,subsequently promoting the proliferation and migration of pancre-atic cancer cells.关键词
