Curcumin induces ferroptosis in hepatocellular carcinoma cells via the Nrf2/HO-1 signaling pathway

Abstract
Objective To explore the effect of curcumin on ferroptosis in hepatocellular carcinoma cells and elucidate its mechanism.Methods Human hepatocellular carcinoma PLC cells were treated with different concentrations of curcumin(0,5,10,25,50,100 μmol/L)for 12,24,and 48 h,respectively,and then the cell viability was evaluated using CCK-8 assay.Human hepatocellular carcinoma PLC cells were divided into four groups:control group,curcumin group,curcumin+ferrostatin-1(Fer-1)group,and cur-cumin+deferoxamine mesylate(DFO)group.The cells in each group were treated with the corresponding drugs at the same time for 24 h,and then CCK-8 assay was used to assess the effects of curcumin and ferroptosis inhibitors(Fer-1 and DFO)on cell viability.Human hepatocellular carcinoma PLC cells were divided into four groups:control group,curcumin group,Fer-1 group,and curcumin+Fer-1 group.After treatment for 24 h,intracellular ROS and Fe2+levels were measured using fluorescent probes DCFH-DA and Ferro-Orange,GSH and MDA levels were quantified using GSH and MDA detection kits,and protein expression levels of Nrf2,HO-1,and GPX4 were analyzed by Western blot.Results The 25 μmol/L curcumin inhibited the cell viability after treatment for 24,48 h(P<0.05),and the 50,100 μmol/L curcumin decreased the cell viability after treatment for 12,24,48 h(P<0.05).Compared with con-trol group,the cell viability was significantly decreased in curcumin group(P<0.01).Compared with curcumin group,the cell viability was increased in curcumin+Fer-1 group and curcumin+DFO group(P<0.05).Compared with control group,intracellular levels of ROS,Fe2+,MDA,Nrf2,and HO-1 were increased while the levels of GSH and GPX4 were reduced in curcumin group(P<0.05).Compared with curcumin group,the levels of ROS,Fe2+,MDA,Nrf2,and HO-1 were decreased while the levels of GSH and GPX4 were increased in curcumin+Fer-1 group(P<0.05).Conclusion Curcumin may induce the ferroptosis by activating Nrf2/HO-1 sig-naling pathway,thereby inhibiting the viability of PLC cells.关键词
