Abstract

Objective To investigate the molecular mechanism of cytokeratin 19(KRT19)regulating cervical cancer cell proliferation through β-catenin signaling.Methods The expression level of KRT19 in cervical cancer and other malignancies was analyzed using the GEPIA online database.HeLa-KRT19(stable KRT19 overexpression)and SiHa-shKRT19(stable KRT19 knockdown)cell lines were established via puromycin selection.Western blotting and immunocytochemistry were employed to detect the expressions of KRT19,β-catenin,and Cyclin D1 in HeLa-KRT19 and SiHa-shKRT19 cells.Cell counting,MTT assays and flow cytometry were used to analyze cell proliferation,cell viability and cell cycle in HeLa-KRT19 and SiHa-shKRT19 cells.Results The results of GEPIA online database analysis revealed significantly elevated KRT19 expression in cervical cancer tissues compared to normal tissues(P<0.05).KRT19 was lowly expressed in HeLa cells but highly expressed in SiHa cells.The stable HeLa-KRT19 and SiHa-shKRT19 cell lines were successfully constructed.Compared with HeLa-GFP cells,β-catenin nuclear translocation was enhanced in HeLa-KRT19 cells,Cyclin D1 expression was upregulated(P<0.05),G0/G1 phase cell percentage was reduced(P<0.05),and the prolifera-tion was increased(P<0.05).Compared with SiHa-GFP cells,β-catenin nuclear translocation was suppressed in SiHa-shKRT19 cells,Cyclin D1 was down regulated(P<0.05),G0/G1 phase cell percentage was increased(P<0.05),and the cell proliferation was decreased(P<0.05).Conclusion KRT19 can upregulate Cyclin D1 protein expression by facilitating β-catenin nuclear translocation,thereby promoting the proliferation of cervical cancer cells in vitro.
