Abstract
Objective To investigate the effects of miR-6892-3p on the proliferation,migration and invasion of cervical cancer cells,and the targeting relationship between miR-6892-3p and SKA2.Methods Thirty cases of cervical cancer tissues from cervical can-cer patients and 30 cases of normal cervical tissues from surgical patients pathologically confirmed benign diseases were selected from Shanxi Bethune Hospital to detect the expression level of miR-6892-3p using RT-qPCR.The expression of miR-6892-3p was measured in cervical cancer cell lines(HeLa,SiHa,CaSki,and C-33A)and normal cervical epithelial cells(HcerEpic),and the cells with the lowest expression of miR-6892-3p were selected for follow-up experiments.The experiment was divided into three groups:control group,mimics-NC group,and miR-6892-3p mimics group.The expression levels of miR-6892-3p and SKA2 mRNA were detected by RT-qPCR,and SKA2 protein expression was analyzed by Western blot.CCK-8 assay was used to assess the cell proliferation capacity,and wound-healing assay and Transwell invasion assays were employed to evaluate cell migration and invasion capabilities.The dual-luciferase reporter assay was conducted to confirm the targeted regulatory effect of miR-6892-3p on the SKA2 gene in cervical cancer cells.Results Compared to normal cervical tissues,the expression level of miR-6892-3p was significantly downregulated in cervical cancer tissues(P<0.01).Similarly,the expression level of miR-6892-3p was significantly lower in the four cervical cancer cell lines than in normal cervical cells(P<0.05),and it was the lowest in SiHa cells.After transfection with miR-6892-3p mimics,the mRNA and protein levels of SKA2 in SiHa cells decreased(P<0.05).Compared to control group and mimics-NC group,the proliferation,mi-gration,and invasion abilities of SiHa cells were reduced in miR-6892-3p mimics group(P<0.05).The dual-luciferase reporter assay indicated that miR-6892-3p significantly inhibited the luciferase activity of the wild-type SKA2 expression vector compared to the mu-tant expression vector(P<0.05).Conclusion miR-6892-3p can exert inhibitory effects on the proliferation,migration,and invasion of cervical cancer cells through targeted regulation of SKA2,and it may serve as a potential therapeutic target for cervical cancer.关键词
