Abstract

Objective To investigate the expression of tripartite motif-containing proteins 32(TRIM32)in cervical cancer patients,and its effect and mechanism on the proliferation,migration and invasion of cervical cancer HeLa cells.Methods Twenty cases of fresh cervical cancer and adjacent normal tissues were collected to detect TRIM32 expression by real-time quantitative fluorescent PCR(qRT-PCR)and Western blot.Wax blocks from cervical cancer and adjacent normal tissues of 85 patients were collected to detect TRIM32 expression by immunohistochemistry.Clinicopathological data of patients were collected to analyze the correlation between TRIM32 expression and clinical features.The expression of TRIM32 in human cervical cancer cell lines HeLa,MS751,SiHa and hu-man normal cervical epithelial cells(HUCEC)by qRT-PCR and Western blot.Human cervical cancer HeLa cells were divided into blank control group,si-NC group group(transfected with an empty vector)and si-TRIM32 group(transfected with a siRNA TRIM32 interference fragment),and the transfection efficiency was verified by qRT-PCR.The proliferation,migration and invasion abilities of HeLa cells were detected by clonal formation assay,scratch healing assay and Transwell assay.The protein expressions of JAK2/STAT3 signaling pathway-related proteins(p-JAK2/JAK2,p-STAT3/STAT3,Ki-67 and c-Myc)were detected by Western blot.Results qRT-PCR and Western blot results showed that TRIM32 expression in cervical cancer tissues was significantly higher than that in adjacent normal tissues(P<0.05).TRIM32 expression level in cervical cancer tissues was significantly higher than that in adja-cent normal tissues by immunohistochemistry(P<0.05),and TRIM32 expression was associated with tumor size,FIGO staging and lymph node metastasis(P<0.05).Compared with HUCEC cells,TRIM32 expression levels in HeLa,MS751 and SiHa cells were in-creased(P<0.05).The expression of TRIM32 in si-TRIM32 group was significantly lower than that in control group and si-NC group(P<0.05),indicating successful transfection.Compared with control group and si-NC group,the proliferation,migration,and invasive abilities of HeLa cells were significantly reduced in si-TRIM32 group(P<0.05),and the protein expressions of p-JAK2/JAK2,p-STAT3/STAT3,Ki-67 and c-Myc were significantly decreased(P<0.05).Conclusion TRIM32 is highly expressed in cervical cancer,and TRIM32 knockdown can inhibit the proliferation,migration and invasion of HeLa cells,which may be related to the inhibition of the progression of JAK2/STAT3 signaling pathway.
