靶向CD74单抗增强宫颈癌新辅助化疗效果分析
Analysis of targeted CD74 monoclonal antibody enhancing the efficacy of neoadjuvant chemotherapy for cervical cancer

Abstract
Objective:To investigate the mechanism by which chemotherapy drug(cis-platin)affects the efficacy of cisplatin in cervical cancer cells by inducing CD74 expression in tumor cells.Method:Cervical cancer specimens were collected clinically for single-cell sequen-cing and immunohistochemical staining.Single cell RNA sequencing was performed on 9 sam-ples from 3 patients.After data quality control,the sequencing data was analyzed,including di-mensionality reduction annotation,reclustering of tumor cell subpopulations,stemness analysis of tumor cell subpopulations,and pseudo temporal analysis.Immunohistochemical staining was used to detect CD74 expression in the samples,and tumor proliferation,migration,invasion,and apoptosis experiments were conducted to verify the biological functional changes of the two groups.Result:Our single-cell sequencing results divided the sample into 7 cell clusters,name-ly tumor cells,mesenchymal stem cells,fibroblasts,endothelial cells,T cells,myeloid cells,and B cells.We performed dimensionality reduction clustering on tumor cells and divided them into 16 tumor subgroups.We analyzed the distribution and expression of CD74 in 7 cell clusters and found that CD74 expression was significantly upregulated in tumor epithelial cells after treat-ment.We collected an additional 50 cervical cancer specimens before and after neoadjuvant chemotherapy treatment,and divided the patients into chemotherapy sensitive group and chemo-therapy resistant group based on their sensitivity to platinum based chemotherapy drugs.The im-munohistochemical results showed that there was no significant statistical significance in the ex-pression level of CD74 in the chemotherapy sensitive group after platinum based chemotherapy drug treatment,while the expression level of CD74 in cervical cancer tissues was significantly increased in the chemotherapy resistant group after platinum based chemotherapy drug treatment(P<0.001).Cell biology experiments suggest that the application of platinum based drugs and inhibition of CD74 expression in tumor cells can reduce tumor invasion,migration,and prolifera-tion ability,and enhance cell apoptosis levels.Conclusion:The expression of CD74 is upregu-lated in tumor cells after NACT treatment,and CD74 targeted antibodies can be used as a sup-plementary drug for neoadjuvant chemotherapy in locally advanced cervical cancer.关键词
