Clinical trials optimize the therapeutic full-process trajectory for advanced metastatic pancreatic cancer
摘要
Abstract
Pancreatic cancer,known as the"king of cancers",shows increasing incidence and mortality rates with insidious onset and highly aggressive nature.Currently,surgery combined with systemic chemotherapy remains the primary approach to prolong survival,yet the 5-year overall survival rate post-operation remains low at approximately 17%.Advanced metastatic pancreatic cancer carries an extremely poor prognosis,with median survival of only 4-6 months and 5-year survival rate below 3%.Although chemotherapy serves as the cornerstone treatment,multi-drug regimens provide limited survival benefits despite improved efficacy,highlighting the urgent need for breakthrough therapies.Recent advances in molecular research and target development have accelerated the transition from traditional to precision treatment approaches for advanced pancreatic cancer,while improved drug accessibility has expanded therapeutic options.However,the complexity of molecular mechanisms and challenges in clinical translation remain key problems to be solved.Research centers worldwide are actively conducting clinical studies on early screening,diagnosis and treatment,demonstrating vigorous development.This review summarizes global clinical research progress on novel therapies for advanced pancreatic cancer,including study designs,mechanisms and targets,as well as new treatment concepts,aiming to improve comprehensive disease management.关键词
