Abstract

Objective:To observe the effects of acupuncture on cognitive function,intestinal mucosal barrier and amyloid β protein(Aβ)in intestine of APP/PS1 mice.Methods:36 male APP/PS1 mice were randomly divided into the model group,the acupuncture group and the probiotic group,with 12 mice in each group.12 male C57BL/6 mice served as the normal control.The acupuncture group was treated with needling Baihui(DU20),Yintang(EX-HN3)and Zusanli(ST36),20 min per time.The probiotics group was treated with probiotics(8.7 × 108 CFU/g).Both groups were intervened once per day for 55 days.Morris water maze was used to observe the behavioral changes.Nissl staining was used to observe the damage of hippocampal neurons.Morphological changes of ileum tissue were observed by HE staining.Immunofluorescence staining was used to observe the expressions of Occludin and Aβ in ileum.The relative expression of Occludin in ileum was examined by Western blotting.Results:Compared with those in the blank group,the escape latency was obviously prolonged(P<0.01);the number of Nissl staining positive cells in hippocampus was reduced(P<0.01);the ileal villus height,the ratio of villus height to crypt height,and the expression of Occludin in ileum were markedly decreased(P<0.01);the expression of Aβ in ileum was significantly increased in the model group(P<0.01).These changes were reversed by acupuncture and probiotics interventions(P<0.01,P<0.05).Conclusion:Needling DU20,EX-HN3 and ST36 can improve the cognitive function of APP/PS1 mice,which may be related to its function in reducing the deposition of Aβ in intestine,alleviating the injury of intestinal mucosal barrier and hippocampal neurons.
