火针调节TRPV1减轻带状疱疹神经性疼痛的作用机制研究
Study on Action Mechanism of Fire Acupuncture in Modulating TRPV1 to Alleviate Herpes Zoster Neuropathic Pain

Abstract
Objective:To observe the action mechanism of fire acupuncture in reducing neuropathic pain of herpes zoster by regulating TRPV1.Methods:In the animal experiment:15 SD rats were randomly divided into the blank group,the model group and the fire acupuncture group,with 5 rats in each group.The VZV-infected herpes zoster neuralgia model was established in the rats.After successful modeling,the model group was left untreated,and the fire acupuncture group was given fire-acupuncture intervention.At the end of the treatment,the right dorsal root ganglion(DRG)neurons were isolated and the expression of TRPV1 in the neurons was detected by Western blot in the three groups.In the cell experiment:primary DRG neurons were isolated and cultured form neonatal healthy SD rats.The cells were randomly divided into the control group,the ET-1 group and the TRPV1 group.Recombinant ET-1 protein was added to mimic neuropathic pain conditions in vivo.In the ET-1 group,cells were treated with recombinant ET-1 protein without further intervention.In the TRPV1 group,after the addition of recombinant ET-1 protein,a TRPV1 antagonist was subsequently added for intervention.The expression of TRPV1 was assessed using Western blot analysis in each group.Results:In the animal experiment:compared with that in the blank group,the expression of TRPV 1 in the DRG neurons was significantly increased in the model group(P<0.05),which indicated high expression of TRPV 1 in the pain model;compared with that in the model group,the expression of TRPV 1 in the DRG neurons was significantly decreased in the fire acupuncture group(P<0.05),which suggested that fire acupuncture treatment could reduce the expression of TRPV1 in DRG neurons.In the cellular experiment:compared with that in the blank group,the expression of TRPV1 in the DRG neuron cells was significantly increased in the ET-1 group(P<0.05),which indicated that TRPV1 existed and acted in the pain model;compared with that in the ET-1 group,the expression of TRPV 1 in the DRG neuron cells was significantly decreased in the TRPV1 group(P<0.05),which suggested that TRPV1 antagonist could reduce the expression of TRPV1 in the DRG neuron cells.Both animal and cell experiments demonstrated that fire acupuncture and TRPV1 antagonists reduced the expression of TRPV1 in neuropathic pain models,which suggested that fire acupuncture could exert an analgesic effect similar to that of TRPV1 antagonists.Conclusion:Varicella-zoster virus(VZV)induces high expression of TRPV1 in rat DRG neurons.Fire acupuncture can reduce TRPV1 expression,thereby producing an analgesic effect similar to that of TRPV1 antagonists.The mechanism by which fire acupuncture alleviates herpes zoster-related neuropathic pain may involve the inhibition of TRPV 1 activity.关键词
