摘要

Abstract

The single factor and orthogonal experiments were used to optimize the composition and content of liquid culture medium for Bacil-lus velezensis 20-10,the optimal culture medium components were maltose 20.0 g/L,starch 10.0 g/L,yeast paste 12.5 g/L,NaCl 1.0 g/L,(NH4)2 SO4 3.0 g/L.In this culture medium,the bacteria concentration was 45.5×108 CFU/mL,which increased by 106.82%compared to the basic culture medium,after being cultured in strain 20-10 shake flasks for 72 hours.The optimal fermentation parameters obtained by opti-mizing the fermentation conditions of 20-10 through single factor and response surface experiments were inoculation amount 2.0%,temperature 30℃,and pH 7.0.Under these optimized conditions,the bacterial concentrations obtained by expanding the cultivation of the strains in 20 L and 500 L fermenters for 64 hours were 52.0×108 and 160.5×108 CFU/mL,respectively,which were 1.36 and 6.30 times higher than the bas-ic culture medium.
