Abstract

Nitraria sibirica,a typical halophytic shrub belonging to the family Nitrariaceae within the order Sapindales,is primarily distributed in desert regions and inland arid areas of China,such as the Hexi Corridor.This plant exhibits remarkable tolerance to abiotic stresses,including drought and salinity.It is frequently utilized as a pioneer species in grassland ecological restoration and the reclamation of heavily saline-alkali soils.Aquaporins(AQPs)are a class of membrane-integrated proteins that regulate the transmembrane transport of water and other small neutral molecules by forming channels across cell membranes,playing a crucial role in plant stress responses.However,the biological functions of the AQP family in the response of N.sibirica to salt stress have not yet been reported.In this study,we explored the salt tolerance mechanisms of N.sibirica by conducting the first genome-wide systematic identification of the NsAQP gene family based on genomic and transcriptomic data.A total of 24 NsAQP genes were identified,with subcellular localization predictions indicating their predominant distribution in the plasma membrane and vacuolar membrane.Through phylogenetic analysis,these genes were classified into five subfamilies:PIP,TIP,NIP,SIP,and XIP,with members within the same subfamily sharing similar conserved motifs and gene structures.Promoter cis-acting element analysis revealed that the NsAQP gene family contains various regulatory elements associated with abiotic stress responses.Analysis of transcriptomic expression patterns under salt stress further demonstrated differential expression profiles of NsAQP genes across different tissues and organs,suggesting their potential involvement in salt stress responses through the regulation of water transport.This study provides a significant theoretical foundation and genetic resources for elucidating the salt tolerance mechanisms of N.sibirica,while also offering new approaches for improving plant stress resistance.
