大豆GmNF-YB2基因非生物胁迫诱导表达及蛋白互作分析

Abstract
Nuclear factor-Y(NF-Y)is a class of transcription factors that can play a role in transcriptional regulation of plants in response to abiotic stress.In order to explore the function and molecular regulatory mechanism of GmNF-YB2 in soybean response to abiotic stress,we cloned the GmNF-YB2 from soybean and performed bioinformatic analysis.Also,we detected the expression level of GmNF-YB2 by real-time fluorescent quantitative PCR(qRT-PCR).The interaction of GmNF-YB2 protein with GmNF-YC6 protein and GmNF-YC15 protein was analyzed by yeast two-hybrid.The results showed that 678 bp GmNF-YB2 gene sequence was obtained by PCR using soybean leaf cDNA as template.The results of bioinformatics analysis showed that GmNF-YB2 gene encoded a protein containing 225 amino acids,with a predicted molecular weight of 24.66 kD and an isoelectric point of 6.97,which was an unstable hydrophilic protein and localized in the nucleus.The GmNF-YB2 protein sequence contained a conserved histone folding motif HFM.Phylogenetic analysis of proteins showed that GmNF-YB2 was closely related to poplar PdNF-YB7 and Arabidopsis AtNF-YB2.qRT-PCR results showed that GmNF-YB2 could be induced by drought,salt and cold stresses in soybean seedlings.The expression of GmNF-YB2 was the highest in soybean stem.The results of yeast two-hybrid showed that GmNF-YB2 protein interacted with GmNF-YC6 protein and GmNF-YC15 protein.In conclusion,these results indicate that GmNF-YB2 may participate in the response of soybean to abiotic stress.关键词
