Abstract

In order to establish an efficient soybean plant regeneration platform in Xinjiang,the cotyledon node of the local soybean variety SY-50 was used as the explant in this study.By exploring the disinfection conditions and the use conditions of plant hormones,a soybean plant regeneration system was established.The basis of this regeneration system,the critical concentration of hygromycin for the regeneration induction of the cotyledon node of this soybean variety was screened,and its genetic transformation system was established.The results showed that the efficient sterilization of seedlings could be achieved by treating with 10%NaClO for 4 min,then with 0.1%HgCl2 for 3 min.The induction medium for adventitious buds was MS+0.5 mg·L-1 6-BA+1.0 mg·L-1 thidiazuron,and the induction efficiency was 33.00%±1.73%.The stem elongation medium for adventitious buds was MS basic medium.The rooting induction medium was MS+1.0 mg·L-1 IBA and complete regenerated plants were finally obtained.Based on the soybean adventitious bud organ regeneration system,the optimal hygromycin screening concentration for the regeneration induction of bean cotyledon node was 15 mg·L-1.The hygromycin resistance screening and PCR identification of transgenic plants were carried out,finally transgenic positive plants were obtained.The results of this study can lay a foundation for the establishment of a technical platform for soybean molecular breeding mediated by the genetic transformation system in Xinjiang,and broaden the range of soybean genotypes improved by molecular breeding technology.
