Abstract

CYP86A subfamily genes are involved in the regulation of plant growth and development,especially in the process of plant response to stress.This study identified the homologous gene GmHORSTs of AtHORST/AtCYP86A1 in soybean by homology comparison.Subsequently,bioinformatics methods were used to comprehensively analyzed the gene structure and cis-acting elements in the promoter region of GmHORSTs,as well as the physicochemical properties and secondary structure of the proteins they encode.The tissue expression pattern of GmHORSTs and the expression changes under NaCl stress were analyzed by qRT-PCR.The subcellular localization of the protein encoded by GmHORSTs in tobacco was analyzed by transient transfection technology.The results showed that soybean CYP86A subfamily genes have eight members,and GmHORSTa(Glyma.11G175900)and GmHORSTb(Glyma.14G192500)are homologous genes of AtHORST/AtCYP86A1 in soybean.The gene structure of GmHORSTa and GmHORSTb is highly similar,the proteins encoded by the genes have the characteristic structural loci of CYP450 protein,and the promoter sequences of the two genes contain a variety of cis-acting elements related to growth and development,hormone response and stress response,and the physicochemical properties of the proteins are obviously different,but the secondary structural components are the same and both are transmembrane proteins without signal peptides.The qRT-PCR analysis revealed that GmHORSTa and GmHORSTb are root-specifically expressed genes and are upregulated by NaCl stress induction.Subcellular localization results demonstrated that both GmHORSTa and GmHORSTb proteins are localized to the endoplasmic reticulum.This study provides a reference for further analyzing the function of GmHORSTs in the process of salt stress response.
