Abstract

In order to screen for salt tolerant soybean germplasm resources suitable for planting in Heilongjiang Province,this study used 333 soybean germplasm resources from 24 countries as materials.Firstly,66 materials were randomly selected for gradient salt stress(0,60,90,120,and 150 mmol·L-1 NaCl).Then,during the soybean germination period,salt tolerance tests were conducted on 333 soybean seeds using the selected optimal NaCl concentration.The germination potential,germination rate,seedling height,root length,and total weight were measured,and their relative germination potential,germination rate,relative root length,relative seedling length,and relative fresh weight were calculated.The salt tolerance ability of different soybeans was analyzed with comprehensive evaluation using a combination of genus function and comprehensive salt tolerance index analysis.The results showed that 90 mmol·L-1 NaCl was the optimal stress concentration for salt tolerance identification during germination determined the gradient salt stress test of 66 soybean germplasm resources.At this concentration,soybean germination potential and germination rate were significantly reduced by 11.8%and 25.2%compared to the control,and could effectively distinguish differences between germplasms.Based on this,a systematic evaluation of 333 soybean germplasm resources revealed that the coefficients of variation for relative germination potential(RGE)and relative fresh weight(RFW)were the highest(162.8%and 134.8%),which can be used as key indicators for salt tolerance screening.Further analysis showed a highly significant positive correlation(r=0.62)between RGE and RFW,revealing the synergistic response mechanism between seed germination ability and biomass accumulation.By using membership functions and cluster analysis,the germplasm was divided into 5 categories,and 3 highly tolerant germplasm(D value 0.804-0.765)and 154 highly sensitive germplasm(D value 0.334-0.174)were identified.By combining comprehensive indicators with cluster analysis,salt tolerant germplasm can be more accurately screened,providing scientific basis and material foundation for soybean salt tolerant breeding.
